Culture medium Peptone: 10.0, beef powder: 3.0, sodium chloride: 5.0, agar: 15.0,pH: 7.3±0.1(25 ℃)
Subculture procedure 1. Preparation of host bacteria solution: Take BNCC305635(ATCC11303) E. coli and culture them by streaking them on NA plates, grow single colonies after 37 ℃ for 18~24h, pick up single colonies in liquid triangular bottles (NB medium) and culture them in shaking table (37 ℃ 140r/min) until OD600 is between 0.1~0.4 (2~3h) for standby; 2. Phage culture: take BNCC 259107(ATCC11303-B1) E. coli phage T1 freeze-dried powder, add 0.5mLNB to dissolve, and then all of them are injected into the host bacteria solution with OD600 between 0.1 and 0.4, shake well, stand still for 0.5h, and then culture in a shaker (37 ℃ 170r/min) for 24h.
