Culture medium	Base medium (English name: LB): yeast extract 5.0g, peptone 10.0g,NaCl 10.0g, distilled water 1.0L,pH 7.0. Sterilization at 121 ℃ for 15min.
Subculture procedure	(1) Prepare 1 vial of sterile water and 2 90mm resistant plates; ②sterilizing the ampoule, open it in biosafety cabinet, heat the tip of ampoule in a flame, quickly drop sterile water to creak it, then break it with forceps; (3) draw 0.5mL of sterile water into the freeze dried ampoule, fully rehydrate and distribute into the resistant plate well in 200ml/plate; (4) The resistant plate was placed under the above specified conditions for 18-24h and then observed (it is forbidden to seal and wrap the plate); ⑤ Pick a single colony and inoculate it into 50mL of liquid LB resistant medium, and culture in a 37 ℃ shaker overnight; ⑥ According to the experimental requirements, absorb appropriate amount of bacteria liquid for plasmid extraction;
