Bacillus subtilis var. niger spore carrier (steel plate)
Culture medium Tryptone: 17.0, soy peptone: 3.0, sodium chloride: 5.0, dipotassium hydrogen phosphate: 2.5, glucose: 2.5g,pH:7.3±0.2(25 ℃)
Subculture procedure 1. Before disinfection, place the bacteria slices on the disinfection evaluation site according to the distribution requirements; 2. After the disinfection reaches the action time, use sterile forceps to transfer the bacterial slice into a test tube containing 5.0mL of corresponding neutralizing agent, vibrate 80 times in the palm of the hand or mix it with a mixer, and neutralize for 10min. 3. Establish a positive control group. During low-temperature field disinfection, the positive control group and the experimental group were put into the corresponding low-temperature environment, and after reaching the same low temperature, they were put into the diluent. 4. Shake the sampling tube on a mixer for 20s or hit the palm of the hand for 80 times, suck 1.0mL of the sample to be tested and inoculate it on a sterile plate, inoculate 2 plates in parallel for each sample, and use the corresponding culture medium to inoculate and culture according to the standard requirements. 5. Count the number of colonies and calculate the ing rate. In accordance with the "GBT38502-2020 disinfectant laboratory germicidal efficacy test method" instructions for use of environmental disinfection field evaluation;
Growth conditions 30 ℃,18-24h, aerobic
Storage conditions -20 ℃
