Standard quality control samples of heat-resistant coliform bacteria in drinking water
Culture medium Peptone: 10.0, beef powder: 3.0, sodium chloride: 5.0, agar: 15.0,pH: 7.3±0.1(25 ℃)
Subculture procedure Samples required a total of 100mL of sterile water or sterile saline to dissolve. Before use, place the quality control sample at room temperature for 30min, then open it in the biosafety cabinet, immediately add 3~4mL of sterile water, suck it out after dissolution, and put it into a sterile bottle. The remaining sterile water is repeatedly washed with the inner wall of the vial, and all of it is transferred to the above sterile bottle, which is the standard quality control sample, totaling 100mL, with a concentration of 10-100CFU/100mL.
Growth conditions 30 ℃;24-48h; aerobic;
Storage conditions -20 ℃, protected from light
