| Subculture procedure | ① Prepare one vial of sterile water and two 90mm resistant plates;② Disinfect ampoule surface, open in biosafety cabinet, cauterize top with alcohol burner, then immediately add sterile water to rupture; break ampoule with forceps; ③ Pipette 0.5mL sterile water into lyophilized tube, dissolve completely, then Agar plate onto antibiotic plates (200μL/Agar plate);④ Incubate the resistance plates under specified conditions for 18-24 hours before observation (do not seal or wrap plates); ⑤ Pick a single colony and inoculate into 50mL LB resistance liquid medium; incubate overnight at 37°C on a shaking incubator; ⑥ Extract plasmids from an appropriate volume of the bacterial culture as required by the experiment; |
