Description LB Medium with 50μg/mL Ampicillin
Composition Yeast Extract:5.0g, Peptone:10.0g, NaCl:10.0g, pH:7.0(25℃)
Growth conditions 37°C; 18-24h; aerobic
Subculture procedure ① Prepare 1 vial of sterile water and 2 90mm resistant plates;② Disinfect ampoule surface, open in safety cabinet, cauterize top with alcohol lamp, then immediately add sterile water to rupture. Break ampoule with forceps; ③ Pipette 0.5 mL sterile water into lyophilized tube. After complete dissolution, spread solution onto antibiotic plates (200 μL/Agar plate);④ Incubate the resistance plates under specified conditions for 18-24 hours before observation (do not seal or wrap plates); ⑤ Pick a single colony and inoculate into 50mL LB resistance liquid medium; incubate overnight at 37°C on a shaking incubator; ⑥ Extract plasmids from an appropriate volume of the bacterial culture as required by the experiment;
