| Subculture procedure | ① Prepare 1 vial of sterile water and 2 90-mm resistance plates; ② Disinfect the surface of the ampoule, open it in a safety cabinet, flame the top with a酒精灯, then quickly add a drop of sterile water to break it open, and subsequently crush it with tweezers; ③ Pipette 0.5 mL of sterile water into the lyophilized vial; after complete dissolution, streak the solution onto the resistance plates at 200 μL per Agar plate; ④ Incubate the resistance plates under the specified conditions for 18–24 h before observation (do not seal or wrap the plates); ⑤ Pick a single colony and inoculate it into 50 mL of liquid LB resistance medium; incubate overnight on a shaking incubator at 37°C; ⑥ Depending on experimental requirements, pipette an appropriate volume of the bacterial culture for plasmid extraction; |
