Subculture procedure ① Prepare 1-2 pieces of the aforementioned agar plates (pre-incubated in an anaerobic environment for 24 hours to remove oxygen); ② Open them in a safety cabinet, ignite the top with an alcohol burner, quickly drop sterile water to rupture them, and then use tweezers to break them into pieces; ③ Pipette 0.5mL of sterile water into the lyophilized tubes, fully dissolve the bacterial powder, and then dispense 200μL per Agar plate, spreading it evenly; ④ Incubate the agar plates under the aforementioned culture conditions until the bacterial strain grows, and then they are ready for use.

If it is overdue, it is deemed that the bacteria are well. The freeze dried culture shall be used up once and shall not be retained. The bacterial viability will resume after 1-2 generation of recovery and can be used normally. Handling instructions of agar plate is enclosed. Please operate in strict accordance with this instruction, otherwise the replacement of bacteria are not be available in case of aberrant growth and loss of viability.

Growth conditions: 37°C, strictly anaerobic, Columbia blood plate (ready-to-use), 3-4 days.

Recovery steps:

① Prepare 2 of the above plates;

②Open it in the biosafety cabinet, sterilize of the ampoule, heat the tip in a flame, quickly drop sterile water to creak it, then break it with forceps;

③Draw 0.5ml of sterile water(Placed in anaerobic environment for 24h equilibrium) into the freeze dried ampoule, make it fully dissolved, and distribute the solution to two plates well in 200ul/plate;

④Put the plates under the above culture conditions for cultivation 3-4days.

Item test results
viability: good viability, in 3days strain layer become obvious;colony is typical on the streaked plate
colony morphology: (above)

size: 1-2mm ; Shape: round ;Edge: irregular ; Transparency: opaque
