| Subculture procedure | ① Thaw Frozen vials in water bath (fungi 30°C, bacteria 37°C), dissolve by rapid shaking; ② Disinfect outer surface of Frozen vials with alcohol swab, then transfer to biosafety cabinet for handling; ③ Unscrew cap, aspirate entire thawed contents, and Agar plate onto 1–2 agar plates (approx. 200μL per Agar plate; for anaerobic bacteria, pre-deoxygenate medium in anaerobic conditions for 24h); ④ Spread evenly and incubate under specified conditions. |
