| Culture medium | BNCC352241 |
| Description | Columbia blood agar plate |
| Composition | Special Peptone:23.0g, Soluble Starch:1.0g, NaCl:5.0g, Agar:10.0g, Defibrinated Sheep Blood:5%(added at 50℃), pH:7.3±0.2(25℃) |
| Growth conditions | 37°C; 3-4 days; anaerobic; |
| Subculture procedure | ① Prepare 1-2 fresh plates as described above (pre-deoxygenated in anaerobic environment for 24 hours); ② Open in a biosafety cabinet, cauterize the top with a酒精灯, then quickly add sterile water to rupture it; break it into pieces with forceps; ③ Pipette 0.5mL sterile water (pre-deoxygenated in anaerobic conditions for 24h) into the lyophilized tube, dissolve completely and mix thoroughly; ④ Pipette 0.2 mL of the bacterial suspension onto a Agar plate and spread evenly. Repeat twice to obtain two plates; ⑤ Incubate under the specified conditions. The culture is ready for use once the strain grows. |
