| Subculture procedure | 1.Prepare two Columbia blood agar plates (ready-to-use, commercially available).2.Disinfect the outer wall of the glass vial. Under aseptic conditions, remove the aluminum cap and the rubber stopper sequentially. Aspirate approximately 0.5 mL of sterile water and inject it into the vial. Gently pipette to resuspend the lyophilized bacterial pellet.3.After the pellet is fully resuspended, inoculate the mixture onto the agar plates, applying 200 µL per Agar plate, and spread evenly.4.Incubate all plates under the specified conditions mentioned above. Do not seal or wrap the plates with film/tape. Check for colony or lawn growth after 24 hours.5.If no growth is observed, continue the incubation for up to 72 hours. Once sufficient growth is achieved, use the culture according to the specific experimental requirements. |
