Subculture procedure ① Prepare one test tube containing 5-10 mL of liquid culture medium and two plates; ② Open the safety cabinet, burn the top with an alcohol lamp, quickly drip sterile water to break it, and then use tweezers to crush it; ③ Transfer 0.5mL of liquid culture medium into a freeze-drying tube, dissolve it thoroughly, and then transfer it back into the liquid test tube, mixing well; ④ Transfer 0.2mL of bacterial suspension into a Agar plate, apply evenly, and repeat twice to obtain two plates; ⑤ Place all liquid test tubes and plates under the above cultivation conditions, and the bacterial strains can be used once they grow.

Medium : liquid culture medium thioglycolate liquid culture medium (formula as follows); The solid culture medium is Columbia blood plate (ready-to-use finished product); Thioglycolate: 15.0g of caseptone (trypsin hydrolysis), 5.0g of yeast extract, 5.0g of glucose, 0.5g of sodium thioglycolate, 0.5g of L-cystine, 2.5g of sodium chloride and 0.001g of azure, agar 0.75g, distilled water 1.0L,pH 7.1 ± 0.2. Sterilization at 121 ℃ for 15min.

Recovery steps:

(1)Prepare a φ18mm sterile test tube of 25ml liquid medium; Silica gel plug is used for the liquid test tube;

(2)Sterilize the ampoule, use a small grinding wheel to make a circle at 1/2 of the tube body under strict aseptic conditions,, and then break it

(3)Draw 0.5ml of liquid growth medium into a freeze dried ampoule, make it full dissolved;

(4)Transfer all the solution to the liquid test tube, plug the tube with silica gel.

(5)Place the liquid test tube in incubator under the above growth conditions for cultivation, The culture solution is obviously turbid or precipitated, and this indicates the bacterial grows well.
