| Subculture procedure | ① Prepare 1-2 plates as above; ② Open plates in a biosafety cabinet after surface disinfection; ③ Pipette 1-3 ml sterile water onto plates; use a sterile spreader to scrape bacterial film from surface to create suspension; ④ Pipette suspension with Pasteur pipette onto fresh plates and spread evenly; ⑤ Incubate plates under specified conditions. (Keep plates moist during incubation. If plates dry out, open them in a safety cabinet and add appropriate amounts of tryptic soy broth.) |
