| Culture medium | BNCC352241 |
| Description | Columbia blood agar plate |
| Composition | Special Peptone:23.0g, Soluble Starch:1.0g, NaCl:5.0g, Agar:10.0g, Defibrinated Sheep Blood:5%(added at 50℃), pH:7.3±0.2(25℃) |
| Growth conditions | 37°C; 24–48 h; anaerobic; (modified PYG liquid medium) |
| Subculture procedure | ① Prepare 1–2 fresh plates as described above (pre-incubated in an anaerobic environment for 24 hours); ② Open in a safety cabinet, scorch the top with a酒精灯, then quickly add a drop of sterile water to crack it open, and subsequently crush it with forceps; ③ Pipette 0.5 mL of liquid medium or sterile water (previously placed in an anaerobic environment for 24 hours to remove oxygen) into the freeze-dried vial; dissolve thoroughly and mix well; ④ Pipette 0.2 mL of the bacterial suspension onto the agar plate and spread evenly; repeat twice to obtain two plates; ⑤ Incubate under the aforementioned conditions; the strain is ready for use once colonies appear. |
| Storage conditions | 2–8°C |
| Safety level | 1 |
| morphology | Colonies are 0.5–1 mm in diameter, round, with smooth, well-defined edges; they are opaque, white on the upper surface, convex in the center, and have a smooth surface; G+ (blue-violet); bacilli; purity: pure |
