Description Nutrient Agar
Composition Peptone:10.0g, Beef Extract Powder:3.0g, NaCl:5.0g, Agar:15.0g, pH:7.3±0.1(25℃)
Growth conditions 28°C; 24–48 h; aerobic;
Subculture procedure ① Prepare 1–2 of the above agar plates; ② Open in a safety cabinet, scorch the top with a Bunsen burner, then quickly add a drop of sterile water to rupture it, and subsequently crush it with forceps; ③ Pipette 0.5 mL of sterile water into a freeze-dried vial, dissolve the bacterial powder thoroughly, then pipette approximately 200 μL onto each of the two plates and spread evenly; ④ Incubate the plates under the specified conditions; the culture is ready for use once colonies appear.
