Description Buffered Charcoal Yeast Extract Agar
Composition Yeast Extract Powder:10.0g, Activated Charcoal:2.0g, Agar:13.0g, α-Ketoglutarate Monopotassium Salt(C5H5KO5):1.0g, ACES Buffer:10.0g, Potassium Hydroxide(KOH):2.8g, Ferric Pyrophosphate(Fe4(P2O7)3):0.25g
Growth conditions 37°C; 24-48h; aerobic;
Subculture procedure ① Prepare 1-2 plates as described above; ② Disinfect ampoule surface, open in safety cabinet, cauterize top with alcohol lamp, then rapidly add sterile water to rupture. Crush contents with forceps; ③ Pipette 0.5mL sterile water into lyophilized tube. After complete dissolution, dispense 200μL/well onto plates and spread evenly; ④ Incubate plates under specified conditions until colonies emerge.
