| Subculture procedure | ① Prepare 1-2 plates as above (pre-deoxygenated in anaerobic environment for 24h); ② Open in a biosafety cabinet, cauterize the top with an alcohol lamp, then quickly add sterile water to rupture it. Break it into pieces using forceps; ③ Pipette 0.5mL sterile water (pre-deoxygenated in anaerobic conditions for 24h) into the freeze-dried vial. After fully dissolving the bacterial powder, dispense 200μL/Agar plate and spread evenly; ④ Incubate plates under the specified conditions. The culture is ready for use once colonies appear. |
