Composition Tryptone:10.0g, Beef Infusion Powder:10.0g, Yeast Extract Powder:5.0g, Malt Extract Powder:150.0g, Glucose:20.0g, Ammonium Citrate Dibasic(C6H8O7.(NH4)2):2.0g, CH3COONa:5.0g, K2HPO4:2.0g, MnSO4:0.19g, MgSO4.7H2O:0.58g, Tween 80:1.0mL, C3H7NO2S.HCl:1.0g, Acetic Acid:20.0mL, Agar:18.0g
Growth conditions 37°C; 7-14 days; anaerobic
Subculture procedure ① Prepare 1-2 fresh plates as described above (pre-deoxygenated in anaerobic conditions for 24 hours); ② Open in a safety cabinet, cauterize the top with an alcohol lamp, then quickly add sterile water to break it. Use forceps to crush it; ③ Pipette 0.5 mL of liquid medium or sterile water (pre-deoxygenated in anaerobic conditions for 24 hours) into the freeze-dried vial. Dissolve completely and mix thoroughly; ④ Pipette 0.2 mL of the bacterial suspension into the Agar plate and spread evenly. Repeat twice to obtain two plates; ⑤ Incubate under the specified conditions. The culture is ready for use once the strain grows.
