Medium : comprehensive PDA agar medium
potato boiling solution 1.0L, glucose 20.0g,KH2PO4 3.0g,MgSO4·7H2O 1.5g, vitamin B1 trace, agar 20.0g (without liquid medium), pH 6.0±0.2. Sterilization at 121 ℃ for 15min. Potato boiling liquid: weigh 200g of peeled potato pieces, boil in boiling water for 30min, and collect the filtrate to a constant volume of 1.0L.
Recovery steps:
(1)Prepare a sterile test tube of 10ml liquid medium and 2 pieces of 90mm agar plates;
(2)Sterilizing of the ampoule, under aseptic conditions, remove the cap and take out the inner vial. lift the cotton plug, rehydrate the pellets by drawing 0.5ml of liquid culture medium into the vial.
(3)Fully dissolved, transfer the solution to the liquid test tube, mix evenly;
(4)Inoculate a plate with the solution, distribute it well in 200ul/ plate; the plates are not allowed to seal by wrapping.
(5)Put all the plates and test tubes under the above culture conditions for cultivation, The culture solution is obviously turbid after 18-24 hours, and this indicates the bacterial grow well.
(6)If the strain does not grow, it should be continued to be cultured for 72h, and then used according to the test requirements.
