Description Reinforced Clostridial Medium (RCM)
Composition Yeast Extract:3.0g, Beef Infusion Powder:10.0g, Glucose:5.0g, Pancreatic Digest of Casein:10.0g, NaCl:5.0g, Soluble Starch:1.0g, L-Cysteine Hydrochloride(C3H7NO2S.HCl):0.5g, CH3COONa:3.0g, Agar:0.5g, pH:6.8±0.1(25℃)
Growth conditions 37 ℃; 24-48 hours; anaerobic
Subculture procedure ① Prepare one 18mm test tube liquid medium, about 10mL (placed in anaerobic environment for deoxygenation for 24 hours in advance); ② Dissolve the Frozen vials in a 37 ℃ water bath and shake rapidly for dissolution; Wipe the outer wall of the Frozen vial with an alcohol swab for disinfection, and then transfer it to an anaerobic glove box for operation; ③ Unscrew the cap of the tube, aspirate all the dissolved solution, and transfer the bacterial solution to a new liquid culture medium using a pipette or suction tube in an anaerobic environment; ④ Incubate the liquid test tube under the above cultivation conditions until the bacterial solution becomes turbid and ready for use.
