Subculture procedure ① Prepare two plates as described above; ② Disinfect ampoule surface, open in safety cabinet, cauterize top with alcohol lamp, then promptly add sterile water to rupture. Crush with forceps; ③ Pipette 0.5 mL sterile water into lyophilized tube. Thoroughly dissolve powder, then inoculate two plates (200 μL/Agar plate) with uniform spread; ④ Incubate plates under specified conditions until colonies emerge.

Growth conditions:37 ℃, aerobic, 3d, MRS medium: peptone 10.0g, beef paste 10.0g, yeast powder 4.0g, glucose 20.0g, magnesium sulfate 0.2g, sodium acetate 5.0g, triammonium citrate 2.0g, dipotassium hydrogen phosphate 2.0g, manganese sulfate 0.04g, Tween 80 1.0g, agar 20.0g (not included in liquid medium), distilled water 1.0l. pH 5.7±0.2. Sterilization at 121 ℃ for 15min.

Recovery steps:① prepare a test tube containing 5~10mL of liquid culture medium; (2) Open it in the safety cabinet, burn the top with an alcohol lamp, quickly drop sterile water to break it, and then break it with tweezers; (3) Absorb 0.5mL of liquid culture medium into a freeze-drying tube, fully dissolve and return to the liquid test tube, and mix well; ④ Place the liquid test tube under the above culture conditions, and the strain can be used when it grows out.
