Fecal coliform positive control samples (membrane method)

Culture medium	Nutritional gravy medium (English name: NA/NB): beef paste 3.0g, peptone 10.0g,NaCl 5.0g, agar 20.0g (not included in liquid medium), distilled water 1.0L,pH 7.0. Sterilization at 121 ℃ for 15min. [Note] Adding 5 mgMnSO4 · H2O to culture Bacillus is beneficial to produce spores.
Subculture procedure	Samples require a total of 500mL of sterile water or sterile saline to dissolve. Before use, the quality control sample is placed at room temperature for 30min, then opened in the biosafety cabinet, 3~4mL of sterile water is immediately added, dissolved and sucked out into a sterile bottle, the remaining sterile water is repeatedly washed into the inner wall of the Xilin bottle, and all of them are transferred to the above sterile bottle, which is a positive quality control sample with a total concentration of 500mL and a concentration of 40 ~ 600CFU/L.

3. Instructions

Dissolve the material by 500mL of sterile water or sterile saline. Before use, the quality control sample is placed at room temperature for 30min, then opened in the biosafety cabinet, 3-4ml of sterile water is immediately added, dissolved and transferred into a sterile bottle; rinse the inner wall of the penicillin bottle with the remaining sterile water, and transfer all the solution to the former sterile bottle. It is the positive quality control sample, with a total of 500mL and a concentration of 40~600CFU/L

2. Please handle in a biosafety cabinet under sterile conditions. The material shall be used up once opened.
