Subculture procedure | Samples required a total of 100mL of sterile water or sterile saline to dissolve. Before use, place the quality control sample at room temperature for 30min, then open it in the biosafety cabinet, immediately add 3~4mL of sterile water, suck it out after dissolution, and put it into a sterile bottle. The remaining sterile water is repeatedly washed with the inner wall of the vial, and all of it is transferred to the above sterile bottle, which is the standard quality control sample, equivalent to 100mL of water sample to be tested. It is suggested that if the filtration volume is 100mL, the number of fecal coliform colonies is 20-60, and the total number is less than 200. |
